农杆菌介导的苜蓿细胞基因转化

摘要:

本文采用叶盘法,利用pBI121质粒转化的农杆菌(Agrobacteri-umtumetaciens)LBA4404菌株转化了紫花苜蓿(Medicago sativa L.)高分化系MX1和MX2叶片,用荧光法检测了转化后培养34—108天不同生长时期的愈伤组织中GUS基因的表达.结果表明所有被检测样品均有GUS基因的表达,转化效率高,结果稳定.转化后108天愈伤组织的β-葡萄糖醛酸苷酶(GUS)的活性为最高,达48.075nmol/min·mgFW.是对照的12倍,并且GUS基因的表达强度并未随培养时间增长而减弱.本文结果表明,LBA4404菌株对新疆苜蓿的转化效率和转化方法是理想的,转化愈伤组织正在分化培养,GUS基因在转化再生植株中的表达,包括分子杂交、转录水平和蛋白质表达有待于进一步测定.
